I have a chair that has no stain but has a clear polyurethane over natural oak. How difficult is it to remove poly, apply a dark stain (mahogany), then a coat of poly? Thanks for your opinions/advice.

Not difficult at all.  Get some KleanStrip Premium stripper and follow the directions.  The poly will come off easily in 30 minutes or less.  Neutralize, lightly sand if needed, and then you're ready to stain.
