I have a 5" wide, by 5" deep, by 34" long area of concrete, that was poured into a foundation wall, to support a new door installation. How long do I have to wait before I can drill and install 3/16" Tapcons?

How stressed will the screws be? If much stress, the longer the better. If not a lot a week should be plenty.
Pay attention to how hard it is when you're drilling the hole. If it drills easily it may not hold the screw well.

They're Tapcons. It may not hold the screw well no matter how long you wait.

I would not use tapcons. I would get some nails and drill a slightly undersize hole, then hammer the nails in. They won't pull out. If you also use construction adhesive, the door sill will never move. I know this from experience.

When the concrete turns white you can drill and install the fasteners. Usually 24 hrs or less.
